生物谷报道:在过去的十年里,芯片技术已经在基因表达研究中起到了举足轻重的作用,带来了深广的影响。基因芯片得到海量数据,但这些海量数据是否准确?如何最大限度提高芯片数据的准确性,是科学家们最头疼的问题。尤其是但是随着利用不同芯片平台获得相异甚至大相径庭的结果逐渐增多,许多人产生了对这一技术可靠性的质疑争论。芯片质量控制(MicroArray Quality Control,MAQC)计划因而得以诞生,作为一个标准将对相关数据分析和争论进行评定。
最新著名的基因芯片专家,华人学者石乐明(音译)以同一期Nature biotech 上发表了5篇文章系统阐述有关基因芯片质量控制的问题解决。
外源RNA对照(External RNA controls,ERCs)是MAQC的一个重要部分,对于芯片分析性能测评也是非常重要的一个指标,但是在各研究团体中并未能得到充分利用。在这一研究报告中,研究人员对两类ERCs进行了评估:其中一类在样品扩增和标记之间加入到总RNA中,另外一类怎是在杂交之前加入到copyRNAs (cRNAs)中,通过多商业芯片平台(multiple commercial microarray platforms获得了ERC浓度反应曲线(concentration-response curves),辨认出在分析过程中出现问题的分析以及潜在的变量来源,除此之外研究人员也对不同的ERC类型的行为进行了检测。这些ERC评估将为未来的多平台分析性能评估中ERC的更好使用提供了一个基础。在这一研究中,浙江大学部分研究工作者参与了这一计划。

Dr. Shi co-authored about 100 peer-reviewed publications in chemoinformatics, bioinformatics, and data mining.
