动脉粥样硬化是已被公认的脑卒中独立危险因素之一,其发生与血脂代谢有着极为密切的联系。人们推测高水平的B族I型清道夫受体在介导高密度脂蛋白的抗动脉粥样硬化发生方面可能具有很重要的作用。目前已有的对B族I型清道夫受体基因多态性的研究主要是针对外显子1 G4A多态和外显子8 C1050T多态,其中外显子1 G4A多态对血脂的影响获得较多的支持,该多态位点不仅对部分人种健康人群血脂有一定的影响,还参与了糖尿病患者血脂代谢紊乱。《中国神经再生研究(英文版)》杂志2013年6月第16期发表的一项关于“Susceptibility gene for stroke or cerebral infarction in the Han population in Hunan Province of China”的研究证实,动脉粥样硬化性脑梗死患者外周血B族I型清道夫受体基因外显子1 G4A的A等位基因频率明显低于正常对照者。但B族I型清道夫受体外显子1 G4A GA+AA基因型的动脉粥样硬化性脑梗死和脑出血患者外周血高密度脂蛋白胆固醇水平均显著高于GG基因型患者,而低密度脂蛋白胆固醇水平均显著低于GG基因型患者。作为认为,B族I型清道夫受体基因外显子1 G4A多态性可能与中国湖南地区汉族人群脑出血无关,而与动脉粥样硬化性脑梗死有关,且A等位基因可能与高密度脂蛋白和低密度脂蛋白代谢有关。(生物谷 Bioon.com)

Susceptibility gene for stroke or cerebral infarctionin the Han population in Hunan Province of China

The scavenger receptor class B type I gene can protect against atherosclerosis; a mononucleotidepolymorphism is associated with differences in blood lipid metabolism, postprandial serum lipidlevels, insulin resistance, coronary artery disease and familial hyperlipidemia. In this study, thescavenger receptor class B type I gene exon 1 G4A gene polymorphism in atherosclerotic cerebralinfarction patients, cerebral hemorrhage patients and normal controls was detected using thepolymerase chain reaction-restriction fragment length polymorphism method. The results showedthat the GA + AA genotype frequency of scavenger receptor class B type I gene G4A inatherosclerotic cerebral infarction patients was similar to that in cerebral hemorrhage patients andnormal controls; however, the A allele frequency was significantly lower than that in normal controls.The serum level of high-density lipoprotein cholesterol in patients with the scavenger receptor classB type I gene G4A GA + AA genotype was significantly higher, while the serum level of low-densitylipoprotein cholesterol was significantly lower than that in patients with the GG genotype, in both theatherosclerotic cerebral infarction and cerebral hemorrhage groups. The serum level of high-densitylipoprotein cholesterol in patients with the scavenger receptor class B type I gene G4A GA + AAgenotype was significantly higher, while the serum levels of low-density lipoprotein cholesterol andtotal cholesterol were significantly lower than those in normal controls with the GG genotype. Ourexperimental results suggest that the G4A polymorphism of the scavenger receptor class B type Igene is a possible predisposing risk factor for atherosclerotic cerebral infarction, and that it has noassociation with cerebral hemorrhage in the Han population in Hunan province of China. The Aallele is possibly associated with the metabolism of high-density and low-density lipoproteincholesterol.
