Few disciplines in medical research have offered as much hope for success and received as much criticism for failure as the field of gene therapy. Equally controversial is a technique currently being developed that aims to repair genes in situ, using the DNA-repair machinery of the cell. Although still in its youth, this approach might be the ultimate therapy for many inherited disorders. Li Liu and colleagues investigate in the current issue of Nature Reviews Genetics.
